Dendritic cells (DCs) have attracted much interest for their potent capacity to initiate immune responses. Here, we show that fusion of a hen egg white lysozyme (HEL) epitope to antigen-Ig fusion proteins (Fabs) efficiently induces HEL-specific CD4(+) and CD8(+) T cells. The HEL-Ig-Fab fusion protein efficiently stimulated CD4(+) T cells, but not CD8(+) T cells, to produce IFN-gamma, IL-2 and IL-4 in vitro. Conversely, DC pulsed with the HEL-Ig-Fab fusion protein activated CD8(+) T cells and induced the production of IFN-gamma, IL-2 and IL-4. DCs pulsed with the HEL-Ig-Fab fusion protein induced CD4(+) T cells and CD8(+) T cells in vivo, but a majority of the CD8(+) T cells produced IL-4. These results show that the HEL-Ig-Fab fusion protein efficiently stimulates HEL-specific T cells to produce IFN-gamma, IL-2 and IL-4 and that it is possible to induce both TH1 and TH2 cells to efficiently produce cytokines. These results provide a method to use DCs as a vaccine adjuvant to induce an optimal type 1 or 2 response.Q:
